A sampler is one of the most useful tools a music composer can have (a good pair of ears and musical imagination, top this of course!). Most pro recording studios in the world have at least one sampler, usually several.
In its simplest form, you assign sounds (wav or whatever) to notes on a keyboard, then you can play them back just as you would any other keyboard sound. In its most advanced form you could have a whole orchestra of realistic sampled instruments playing, and as long as it was sequenced and mixed well, few people could tell it was fake. (Have a listen to some of the demos from the vienna symphonic sample library)
Its very expensive but a lot cheaper than hiring an orchestra every time you want to hear how your tune will sound with a real orchestra playing it!

a tracking program is pretty much similar to a sampler, you can assign sounds to notes on the keyboard (Range), and you have good control over the way the sounds are played. One good example would be ModPlug tracker, or Renoise, or Skale Tracker.
on top of this you could get a software sampler , like Halion, Kontakt, Sample Tank, EXS24, VSampler or GigaStudio and load up commercial sample CDS in a sequencer like Cubase SX (VST), or Logic audio VST, or any free VST hosts like Buzz Tracker, or PSycle, or Renoise... etc.
There is no predefined programs for "game music", just the talent of the composer with whatever tools they feel comfortable with and good mastering/post production skills to get the music to match the game.
